Isolating nucleic acids from nucleic acid-containing samples, where the samples are mechanically broken down with known methods, comprises adding a chaotropic denaturing agent to the sample, homogenizing the lysed sample and removing the cell debris by centrifugation, adding alcohol to the sample, separating the nucleic acids by binding the nucleic acids to a mineral carrier, washing the nucleic acids, adding sodium citrate or sodium chloride solution to the nucleic acids, and washing and eluting the adsorbed total nucleic acids according to known methods. An independent claim is included for a test kit for isolating nucleic acids from nucleic acid-containing samples, comprising a buffer containing guanidine hydrochloride, guanidine isothiocyanate or guanidine thiocyanate, ethanol, separation column for nucleic acids, washing buffer, sodium citrate or sodium chloride solution and collecting container.
