The present invention relates in a first aspect to polypeptides useful for the prevention or treatment of various diseases. In particular, the present invention relates to a polypeptide containing a homeodomain for the prevention or treatment of lipid-based metabolic diseases, cancer, (neuro)degeneration, autoimmunity, sepsis, cystic fibrosis, diabetes, or for modulating the immune system in an individual or as an anti aging compound. In another aspect, the present invention relates to nucleic acids encoding said polypeptides, vectors containing said nucleic acid molecules as well as host cells comprising said vector. Further, the present invention concerns isolated antisense RNA or DNA molecules. In addition, the present invention provides methods for screening compounds for treating or preventing said diseases based on the ability to alter expression of ceramide synthases and its cleavage.
