Structure and use of 5' phosphate oligonucleotides
Simple SummaryContent extracted from patent full text and abstract with AI.
This invention relates to oligonucleotides (short DNA or RNA molecules) with an uncapped 5' phosphate group, which can stimulate the body's immune defenses by activating molecules such as RIG-I. These oligonucleotides, or similar bacterial RNAs, can be used to induce the production of specific immune signaling proteins (like type I interferon and interleukins), thereby triggering strong anti-viral, anti-bacterial, and anti-tumor responses. The approach can be applied both in laboratory settings and in living organisms, targeting various diseases where immune stimulation is beneficial.
Use CasesContent extracted from patent full text and abstract with AI.
- Treatment of viral infections by boosting the body’s natural antiviral responses.
- Therapy for bacterial infections using immune system activation.
- Cancer immunotherapy to stimulate the immune system to attack tumor cells.
- Management of parasitic infections with enhanced innate immunity.
- Adjunct treatment for allergies by modulating immune signaling.
- Therapeutic support for autoimmune diseases by influencing immune pathways.
- Treatment for immunodeficiencies and immunosuppression to restore immune competence.
BenefitsContent extracted from patent full text and abstract with AI.
- Provides a new method to activate the body’s innate immune system through specific signaling pathways.
- Can be tailored to induce a targeted immune response (e.g., antiviral, antibacterial, anti-tumor).
- Potential to treat a wide range of diseases, including infections, cancers, and immune-related conditions.
- May offer an alternative or adjunct to conventional drugs such as antibiotics and chemotherapy.
- Can be used in personalized medicine approaches for targeted therapy.

Patent Abstract
STRUCTURE AND USE OF 5' PHOSPHATE OLIGONUCLEOTIDES Abstract Oligonucleotides bearing free, uncapped 5' phosphate group(s) are recognized by RIG-I, leading to the induction of type I IFN, IL- 18 and IL- 1 production. Bacterial RNA s also induces type I IFN production. 5' phosphate oligonucleotides and bacterial RNA can be used for inducing an anti-viral response or an anti-bacterial response, in particular, type I IFN and/or IL-18'and/or IL-16 production, in vitro and in vivo and for treating various disorders and diseases such as viral infections, bacterial infections, parasitic infections, tumors, allergies, autoimmune diseases, immunodeficiencies and 10 immunosuppression. Single-stranded 5' triphosphate RNA can be used for inducing an anti-viral response, an anti-bacterial response, or an anti-tumor response, in particular, type I IFN and/or IL- 18 and/or IL- 1 production, in a target cell-specific manner.
